2. Postdoctoral Position

The Conn Lab is looking for 1-2 postdocs focused on post-transcriptional gene regulation and the roles of RNA-binding proteins, RNA modifications, and signaling that are rewired during cancer progression. One project is focused on identifying regulators of selective translation and spatial RNA interactions in the cell during drug resistance in prostate cancer (more mechanism to move into translational research later). The other project is looking at adaptive stress responses in pancreatic tumors and will require mouse modeling, tissue analysis, and treatments in vivo. The Mark Kahn lab in the Perelman School of Medicine (Cardiology) is currently recruiting for a postdoctoral researcher for a grant funded project investigating myxomatous valve disease and the role of flow-responsive signals KLF2 and KLF4 in heart development. The Kahn lab uses complex mouse genetics systems with state-of-the-art biochemical and molecular approaches to understand the novel links between hemodynamic forces and vascular diseases in the arterial and venous system. A Postdoctoral Researcher position is available for a highly motivated applicant in the laboratory of Dr. Fiona Simpkins at the University of Pennsylvania. Our focuses are to understand resistance mechanisms to targeted therapies such as DNA Damage Repair inhibitors (DDRi), to sequence DDRi with Immuno-Oncology (IO) drugs, and to identify biomarkers or response to these agents. We have developed a preclinical drug development program in the Ovarian Cancer Research Center with the goal of moving novel laboratory therapeutic discoveries into the clinic. We have developed >150 patient derived xenografts (PDX) models, patient-derived organoids, and primary tumor cultures from ovarian and endometrial cancers. These PDX models mimic the original tumors, thereby accelerating the translation of preclinical study results into clinical trials for patients.

A postdoctoral position is immediately available in the laboratory of Dr. Alessandra Chesi at the University of Pennsylvania with a focus on discovering and functionally characterizing novel genes for Alzheimer’s, Parkinson’s and other neurodegenerative diseases in human brain relevant cell types. Dr. Chesi has a long-standing interest in genomics and neurodegenerative diseases; she has developed Capture C based approaches to map GWAS signals to their effector genes (see some examples in linked publications) and is applying them to brain-related disorders. Main areas of research in our lab are 3D genomics and epigenomics techniques (such as Hi-C, ATAC-seq, ChIP-seq and RNA-seq), CRISPR/Cas9 editing, pooled CRISPRi/a screens with scRNA-seq readout, plus development of iPSC-based models for neurodegenerative and neurodevelopmental diseases (2D neuronal/glial differentiation, co-cultures, 3D brain organoids).

A postdoctoral researcher position is available in the Department of Psychiatry, University of Pennsylvania. The selected applicant will work under the direct supervision of Dr. Rachel Kember, a computational geneticist with expertise in biomedical informatics and psychiatric disorders. Current projects in the lab focus on understanding the genetic underpinnings of psychiatric and substance use disorders using genetic and phenotypic data from electronic health records with linked biobanks.

Datasets include the Million Veteran Program, a large dataset of over 850,000 veterans with genetic data currently available for 650,000 individuals; the Penn Medicine Biobank, which includes 190,000 patients, 45,000 with genetic data; and the Yale-Penn study, which includes 12,000 individuals with an in-depth substance use and psychiatric assessment. Dr. Kember is co-chair of the PsycheMERGE Diversity Initiative, a collaboration across multiple biobanks formed to address inequity in psychiatric genetics research.

A one year post doctoral fellowship is available immediately with the option to extend an additional 1-3 years, to investigate the role of the extracellular matrix protein type III collagen (Col3) in modulation of the breast cancer microenvironment. Using a combination of in vitro and mouse models, we have initiated multiple lines of investigation that will provide insight into how Col3 inhibits formation of a tumor-permissive microenvironment. This work also capitalizes on our studies using spontaneous naturally occurring large animal models of mammary tumors. Future work will characterize drivers of tumor-permissive and suppressive collagen signatures and investigate the use of a novel tissue engineering strategy to control tumor growth and metastasis following tumor resection to improve clinical outcome in veterinary and human breast cancer patients.

21. Postdoctoral Position 

Summary of Postdoctoral Fellowship:

Two postdoctoral researcher positions are available in the laboratory of Dr. Xiaolu Yang at the Perelman School of Medicine at the University of Pennsylvania. The Yang lab studies cancer and neurodegenerative diseases. Our current projects focus on two broad areas:

(1) The tumor suppressor p53, metabolism, and autophagy. We are interested in the regulation and functions of the preeminent tumor suppressor p53. Our results have revealed a role for p53 in modulating metabolic pathways that are critical for biosynthesis and redox balance. We are investigating the function of p53 as both a sentinel and a regulator for metabolic activities. We are also identifying and characterizing metabolic alterations that drive tumor initiation and progression. A recent extension of this research area is to define the role of metabolism and autophagy in stem cells, including embryonic stem cells and cancer stem cells.

(2) Protein quality control, aging, and neurodegeneration. Our lab recently identified two protein quality control (PQC) systems, which consist of tripartite motif (TRIM) proteins and poly-Asp/Glu (polyD/E) proteins, respectively. Unlike canonical PQC systems, these new systems are independent of ATP and are unique for animals (the TRIM system) or eukaryotes (the polyD/E system). Both systems are multifunctional and highly effective. We are investigating their mechanisms of action; their roles in aging and neurodegenerative diseases, including Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis; and their utility in treating these diseases.
